1336 Use of long-term cultivated human sweat gland cells for generation of spheroids partially mimicking human sweat gland architecture and characteristic marker expression profile

Dysregulated sweat secretion can cause serious clinical problems. Experimentally regenerated human glands (hSWGs) should provide a valuable tool for dissection of hSWG biology and the pathophysiology sweating disturbance, and, ultimately drug discovery disorders; however, full establishment protocols bioengineering requires further endeavour. In this study, we attempted to establish methodology allowing long-termin vitroexpansion cells generation spheroids which mimicking SWG architecture intrinsic marker expression. When hSWGs were micro-dissected from skin samples cultured with or without fibroblast feeder in respective culture medium, success rate primary was higher on-feeder (66.7% vs 42.9%). On cells, cell lines (n = 10) could be passaged 6.9 ± 2.8 times, while feeder-free maintained only 2.6 2.4 passages. both condition, SWG-like colonies noted.
